Technical Tip: Passaging of Human Embryonic Stem Cells
Human embryonic stem cells (hESC) should be routinely passaged as clumps not as single cells to maintain optimal cell quality and survival. hESC passaged as single cells initially show poor survival but with routine single cell passaging, a sub population of cells will be selected which have improved cloning efficiency but will most likely be karyotypically abnormal and are not functional hESC.
We recommend the use of dispase and mechanical methods for passaging hESC grown in mTeSR™1, our medium for the feeder-independent culture of hESC.
